Mensen met een bepaalde variant van het gen KLOTHO hebben betere cognitieve vaardigheden zoals denken, leren en waarneming, ongeacht hun leeftijd of sekse.
Dat schrijven onderzoekers van de University of California San Fransisco uit de Verenigde Staten deze week in het tijdschrift Cell Reports.
Het verhogen van de KLOTHO-niveaus in genetisch aangepaste muizen zorgde bij de diertjes ook voor betere cognitieve vaardigheden.
Het onderzoek kan een belangrijke stap zijn in de zoektocht naar oplossingen voor ouderdomsziekten, zoals Alzheimer en andere dementie-verschijnselen.
Om de cognitieve vaardigheden te testen bij mensen, testten de onderzoekers 718 personen tussen de 52 en 85 jaar. Geen van de onderzochte personen vertoonde tekenen van dementie.
Van de onderzochte personen bezat 26 procent één KLOTHO-gen, KL-VS. De rest was KL-VS-loos. Mensen met twee KL-VS genen (homozygoot) werden uitgesloten van de tests, omdat mensen met dat genotype om onbekende reden een kortere levensverwachting hebben en andere nadelige effecten ondervinden.
Blauwdruk
Het KLOTHO-gen zorgt voor een blauwdruk van een eiwit dat vooral door cellen in de nieren, placenta, dunne darm en prostaat gemaakt wordt. Het eiwit circuleert door het lichaam via de bloedbaan. Mensen met het KL-VS-gen hebben een hogere concentratie van dat eiwit in hun bloed dan mensen met andere varianten.
De mensen met KL-VS-gen presteerden beduidend beter op de cognitieve tests dan de personen die het het zonder moesten stellen, ongeacht of men wel of geen aanleg voor Alzheimer had.
Om uit te vinden of de wetenschappers het bij het rechte eind hadden, testten ze hun idee op muizen die veel klotho-eiwit produceerden. De klotho-muizen leefden langer en hadden hogere concentraties klotho in het bloed en delen van de hersenen.
Doolhof
Ongeacht de leeftijd van de muizen, presteerden de klotho-muizen beter bij het onthouden van de locatie van verstopt voer in een doolhof dan de andere muizen. Ze vonden het voer gemiddeld twee keer zo snel.
Ook bij de muizen bleek dat de effecten van klotho niet leeftijdsafhankelijk zijn. Dit suggereert dat klotho los van het verouderingsproces staat.
De wetenschappers suggereren dat het wellicht mogelijk is de klotho-hoeveelheid bij mensen aan te passen op latere leeftijd, waardoor bepaalde geheugenproblemen zich minder ernstig voordoen.
